De Fraiture holds a BSc in Physical Geography from Utrecht State University in the Nederlands, a MSc in Tropical Land and Water Management from the Wageningen Agricultural University in the Netherlands and a MA in Economics from the University of Colorado in Boulder (USA). The title of her thesis there is “The impact of volumetric water pricing on agricultural water use and farmer profit. A multi-scale modelling framework“.

During her PhD in Civil Engineering, also at the University of Colorado, she specialised on Water Resources Management. The title of her dissertation is “The use of entropy optimization principles in parameter estimation. De Fraiture has over 20 years of field experience in agricultural water management in many different developing countries.

In the beginning of her career, de Fraiture was active in rural development projects in Nepal and Senegal. In the course of her work for the International Water Management Institute from 1996 – 2001 she was based in Colombia, Sri Lanka, Ghana and Burkina Faso. She was Head of the West-Africa Office, Theme Leader in Basin Water Management and Group Head. Since January 2012 she is Professor of Hydraulic Engineering for Land and Water Development at UNESCO-IHE.<ref>http://www.iwmi.cgiar.org/About_IWMI/People/pdf/Charlotte.pdf

De Fraiture also researched in a range of academic projects on [[Watershed_management|watershed development]] and [[Design_for_irrigation|irrigation practices]].

Modelling global water demand and supply in one project she contributed to the progress and launch of the global water and food model (WATERSIM). This model is relevant for the calculation of potential water situations in the framework of the Comprehensive Assessment on Water Management in Agriculture.
